Do you have to drain a fibreglass pool to fix a hole on the bottom or can you use some type of patch in the water?

It might be possible to fix the hole with the water in the pool using Pool Putty epoxy putty. Especially fairly small holes, up to 1" in diameter, this works quite well. If there is a washout behind the hole, you should fill it with something, small gravel, a sponge - anything to fill the void so when you put the epoxy in place it doesn't suck through the hole.
